Researchers Map DNA Quadruplex Pairs Regulating Genes

An international team including HSE University used AI to create the first genome-wide map of DNA G-quadruplexes, retraining DNABERT on EndoQuad to predict roughly 360,000 quadruplex sites. They found these structures often occur as promoter–enhancer pairs that direct tissue-specific gene programs in healthy cells but rewire toward universal growth and division programs in cancer, a result published in Nucleic Acids Research.
Key Points
- 1Predicts roughly 360,000 G-quadruplex sites genome-wide using GQ-DNABERT trained on EndoQuad.
- 2Reveals promoter–enhancer quadruplex pairing that links regulatory elements to tissue-specific gene expression.
- 3Shows cancer cells retain pairs but redirect targets to proliferation programs, informing targeted therapy strategies.
Scoring Rationale
Significant novelty and peer-reviewed publication justify a breakthrough score, though functional validation and therapeutic translation still require further experimental work.
